Determination of Gibberellin Residue in Vegetables and Fruits by High Performance Liquid Chromatography

Abstract

Researched the analytical method for the determination of gibberellin acid residue in vegetables and fruit by high-performance liquid chromatography. The GA3, after being extrated by using80% methoanol water solution, decolored by petroleum ether, extracted by ethyl acetace, and then,purged by reversed extraction with Phosphate Buffer(PB) and extrated with ethyl acetate again, was separated on C18 column using methonl and 0.1% acetic acid solution as mobile phases and 206 nm as HPLC detected wavelength. The samples were qualitatived by retention time and quantified with the external calibration method. The results showed the method performed well within the linear range between 0.25 mg/L and 50 mg/L(r=0.999 9), and the limit of detection was 0.067 mg/kg. The method was used to detect the contents of GA3 in 6 kinds of vegetables and fruit, the recoveries for the spiked samples were ranged from 71.6% to 105.2%, and the variation coefficients were from 2.1% to 9.8%.
