The Effect of Estrogen on the Glomerulosclerosis in SD Rat

Abstract

Objective To explore the effect of estrogen on the glomerulosclerosis. Method Twelve-week-old female Sprague-Dawley rats were randomly allocated to one of 3 treatment groups:1)sham operation; 2)ovarietomy+placebo (ovx); 3)ovariectomy +estrogen(ovx+E2). The rats were assigned to ovariectomy or estrogen treatment groups a week after ovariectomy. Results 1)Serum estradiol concentration were significantly lower in ovariectomized rats than in sham-operated animals. Estrogen replacement therapy led to a recovery of physiological estradiol level. 2) SBP was significantly higher in ovariectomized rats than that of sham-operated rats [(117.5±7.6 vs 104.4±5.2)mm Hg, P 0.05]. No difference was observed between ovariectomized rats with estrogen replacement therapy(ERT) and sham-operated animals. 3)Significant glomerulosclerosis was shown in ovariectomy rats, however estrogen replacement therapy failed to ameliorate the glomerular injury. Conclusion SBP was increased in ovarietomized rats, ERT prevent the increase of SBP. Ovariectomy led to glomerulosclerosis in rats, however estrogen replacement therapy failed to attenuate glomerular injury.
