【Objectives】 To investigate the expression of miR-429 in colorectal carcinoma and normal colorectal mucosa,and explore the role of miR-429 in carcinogenesis and progression of colorectal carcinoma,and to evaluate its clinicopathologic significance.【Methods】 RT-PCR and in situ hybridization(ISH) were performed to detect miR-429 expression in 40 colorectal carcinoma samples and 40 samples from normal colorectal mucosa.The relationship of miR-429 expression and clinicopathologic features were also analyzed.【Results】The results of RT-PCR indicated that the expression of miR-429 is significantly lower in colorectal carcinoma than that in normal colorectal mucosa,and there is significant difference between tumors and normal samples(P 0.05).To verify the results by RT-PCR,we performed the ISH detection,and found that miR-429 is expressed in cytoplasm,the density is decreased in colorectal carcinoma compared to normal colorectal mucosa.Then,the correlation of miR-429 expression levels with clinical characteristics in the 40 CRC patients was analyzed.The low expression of miR-429 is associated with differentiation and malignant progression of colorectal carcinoma.The lower the miR-429 expressed,the less differentiation of colorectal carcinoma,and the difference is significant(P 0.05).At the same time,the expression of miR-429 is lower in the Ⅲ + Ⅳ stage than Ⅰ + Ⅱ stage of colorectal carcinoma(P 0.05).【Conclusions】miR-429 expression is lower in colorectal carcinoma than that in normal colorectal mucosa,and its low expression is closely associated with differentiation and malignant progression of colorectal carcinoma.
