Abstract
Objective To observe the expression of tissue factor(TF) in rat aortic endothelial cells(RAECs) induced by optimal dose of lipopolysaccharides(LPS) for different duration and to find the action time when expression of TF reached the peak level. Methods Cultured RAECs were incubated with LPS at different concentrations of 0.1 μg/ml,1 μg/ml,10 μg/ml and 100 μg/ml, respectively, for 6,12,24,48 hours, cell viability was then determined by MTT, level of TF protein was detected with immunohistochemistry and Western blotting methods. Results LPS at concentrations of 0.1 μg/ml and 1 μg/ml did not produce cell toxicity as shown by cell viability. TF protein level was significantly increased after 1 μg/ml LPS exposure, and reached the peak at about 4h, then began to descend to normal. Conclusion 1 μg/ml LPS promoted the expression of TF protein, and the peak value occured at about 4h.
