Abstract

Objective To study the effects of resveratrol on the proliferation and apoptosis of tongue cancer cell Tca.Methods Tongue cancer cell line Tca was cultured to study the effects of resveratrol on cell proliferation.Firstly,MTT assay was used to evaluate the IC50 of resveratrol on the cell line.Secondly, through colony formation assay, the ability of cell proliferation, when exposed to resveratrol at 75 μmol/L,was detected. At last, the distribution of cell cycle and apoptosis was analysed by flowcytometry,Hoechst33258 staining was used to show the morphology of apoptotic cells. Results Resveratrol could inhibit the growth of Tca cells in varied degrees. The IC50 was about 100 μmol/L and 60 μmol/L after treated by resveratrol for 48 h and 72 h.When the cells were cultured with resveratrol(75 μmol/L) for 48 h,resveratrol could highly inhibit the ability of Tca colony formation, and also caused G1 arrest and apoptosis. Conclusion Resveratrol could inhibit Tca cell proliferation, and cause G1 arrest and apoptosis. It may be a possible new way for the therapy of tongue cancer.
