The effects of triptolide on the proliferation and apoptosis of pancreatic cancer cell

Abstract

Objective:To investigate the effects and the possible mechanism of triptolide on the growth and apoptosis of human pancreatic cancer cell in vitro.Methods:The pancreatic cancer cell line SW1990 was treated with triptolide.MTT method was used to determine the inhibitory effect of triptolide on cell proliferation,and flow cytometry (FCM) with AnnexinⅤ-FITC and propidium iodide (PI) staining was used to determine apoptosis induction and blocking effects of triptolide on the cycle proliferation.Results:After the treatment with triptolide,cell proliferation was blocked by triptolide,which showed a significant positive association with the level of triptolide concentration(P0.01),and the apoptotic rates of SW1990 cells were increased in concentration-dependent and time-dependent manners,and its cell cycle was arrested at G0/G1 phase.Conclusion:Triptolide is able to inhibit proliferation,induce the apoptosis and block cell cycle effect on pancreatic cancer cells.The potential for its treatment is worth further investigating.
