Effects of Kangnaoye 1 on Cerebral Ischemia-reperfusion Half Silent Zone Apoptosis and Bcl-2/Bax Ratio in Rats

Abstract

Objective To observe the effect of Kangnaoye 1 on nervous apoptosis and Bc1-2/Bax ratio in SD rats after cerebral ischemia-reperfusion,to explore its possible protective mechanism for cerebral ischemia-reperfusion.Methods SD rat models of middle cerebral artery occlusion(MCAO) were copied and given 12-,24-,72-hour reperfusion after 2 h of ischemia.One hundred eighty rats were randomized into groups A(sham operation group),B(model group or ischemia-reperfusion group),C(receiving fasudil hydrochloride injection),D(receiving Kangnaoye 1) and E(receiving Kangnaoye 1).The effects of Kangnaoye 1 on cerebral ischemic neurological function,infarct size and pathomorphology.The number of ischemia half silent zone apoptosis and Bc1-2/Bax ratio were determined by TUNEL method and immunohistochemistry.Results The apoptosis number increased in group B as compared with group A.Kangnaoye 1 reduced,in varying degrees,the scores of nervous function,infarct size,the changes of pathomorphology in experimental ischemic rats(P0.05).In groups D,E,the number of cerebral ischemia half silent zone nervous apoptosis decreased,Bc1-2/Bax ratio increased,there was not significant difference between 2 groups(P0.05).Conclusion Kangnaoye 1 can promote the repair and remodeling of cerebral injuries by regulating neuronal apoptosis and Bc102/Bax ratio,thus to play a role in cerebral protection.
