Investigation on the cultivation of rabbit marrow stem cell in vitro

Abstract

Objective To investigate the method of culturing rabbit marrow stem cell(MSC)and observe its growing character.Methods Bone marrow was extracted from the iliac of rabbit.MSC was then separated from the marrow and cultured with DMEM media and DMEM+mineralized liquid respectively.The growth curve,ALP stain and calcification nodule formed by MSC were observed.Results Growth curves of passage 1-3 MSC had the similar characters and when the MSC was cultured with mineralized media.It showed the character of osteoblast,such as positive ratio of ALP stain,increased greatly and could form calcification nodule.Conclusion Rabbit MSCs differentiate into osteoblasts when cultured in vitro,therefore they can be used as the seed cells for the bone tissue engineering.
