Abstract

Objective:To observe the synergistic effect of elemene combined with hyperthermia on cell line A549 of human lung adenocarcinoma.Methods:The sensitivity of the cells treated with gradient concentration and elemene in 42℃ or 37℃ water bath for 1.5 hours were detected by MTT assay.One hour and a half after treatment with water bath at 42℃ combined with 15μg/mL or 60μg/mL of elemene,cell cycle was detected by flow cytometry and appearance of the cells was detected by transmission electron microscope.Results:After hyperthermia,IC50 values of elemene decreased from 35.43μg/mL to 28.68μg/mL.MTT assay showed low concentration elemene combined with hyperthermia had synergistic cell killing effect,and the cells of A549 appeared apoptosis.Apoptotic body and cellular necrosis can be seen.FCM showed elemene combined with hyperthermia had significant effect on cell cycle with increasing G0/G1 phase population and reducing S phase population.Conclusion:Low concentration elemene combined with hyperthermia has synergistic effect of cytotoxicity on A549 cells.
