Effect of Astragalus on Cytokines,the Level of Intracellular Calcium and the Expression of Cyclin D1 gene in Acute Renal Ischemia-Reperfusion Rats

Abstract

Objective:To investigate the effect of astragalus on cytokines in rat model with acute renal ischemia-reperfusion,as well as the changes of the level of intracellular calcium and the expression of Cyclin D1 gene.Methods:Ninety SD male rats were randomly divided into and renal ischemia-reperfusion group(model group),sham operation group(control group),and astragalus group(experimental group).The blood and renal tissue samples were collected respectively at 3,6and 12 hafter reperfusion to determine renal function,cytokines,intracellular calcium level and Cyclin D1 level.Results:Compared with the control group,Cr and BUN of model group increased after 6h,the difference was statistically significant(P0.01);BUN significantly in-creased in experimental group after 6h,and Cr increased after 12 hompared with the control group(P0.05);The difference of Cr between model group and experimental group after 6h was statistically significant(P0.05);so was BUN after 12h(P0.05).IL-6of model group and experimental group at 3,6,12 hafter reperfusion were higher than those in the control group(P0.01),but IL-6in experimental group was lower than that in the model group(P0.01);IL-10 of model group after 6hwere lower than that in the control group and the experimental group(P0.05),there was no significant difference between the other groups(P0.05).TNF-αof experimental group and model group at 3,6,12 hafter reperfusion were higher than in the control group(P0.01),but the TNF-αin experimental group was lower than that in the model group(P0.01).[Ca2+]I of model group and experimental group after 3hwas significantly higher than that of control group(P0.01),but the levels of experimental group at 3hand 6h was lower than those in the model group(P0.01),and there was no significant differences in 12 hbetween model group and experimental group(P0.05);Cyclin D1 of model group after 3hwas significantly higher than that of control group(P0.05),there was difference between experimental group and model group(P0.05)but the levels of experimental group after 6hwas no different significant from those in the model group(P0.05).Conclusion:Astragalus could relieve the ischemic-reperfusion calcium overload of rat,release cytokines,and reduce renal damage.
