Expression of PCNA and nm23 in transitional cell carcinoma of bladder

Abstract

Objective:To investigate the expression of PCNA and to nm23 and to determine their relationship with clinicopathological features in transitional cell carcinoma of bladder.Methods: Immunohistochemical technique,S-P method was used to study the expression PCNA and nm23 in 78 cases transitional cell carcinoma of bladder.Positive level index(PLI)was used to assess expressing level of PCNA and nm23 protein.The percentage of positive cell ,average optical density (AOD) was assessed by image analyzer and positive level index(PLI)=P/A(%)×AOD.Results:Overexpression of PCNA had been observed in transitional cell carcinoma of bladder.Expression PCNA had been observed in all 8 cases normal bladder mucosas.Statistical analysis revealed they had significant difference(P0.01).Lower expression of nm23 had been observed in transitional cell carcinoma of bladder.Expression nm23 had been observed in all 8 cases normal bladder mucosas.Statistical analysis revealed they had significant difference(P0.01).PCNA protein was associated with clinicopathological factors such as T-factor,stage,grade(P0.01);Nm23 protein was not associated with clinicopathological factors such as T-factor,stage,grade in transitional cell carcinoma(P0.05).Both of expression PCNA and nm23 protein were associated with relapse of bladder transitional cell carcinoma,of which PCNA was positively correlated with relapse of carcinoma,(P0.01)and nm23 was negatively correlated with relapse of carcinoma,(P0.01).Conclusions:PLI,as a reflector of immunohistochemical reaction,make the study of immunohistochemistry turn from quality,semi-quantity to quantity and make it easy to be statistically analyzed.Detecting the expression PCNA and nm23 protein in transitional cell carcinoma of bladder has more important meaning to clinical diagnosis and treatment.The expressing level of PCNA can be used to differentiate benign and malignant tumor.Using PLI,by analyzing big sample data,a criteria can be found to diagnose benign and malignant tumor.
