Abstract
Aim To study excisanin A that induced apoptosis in human colon cancer SW620 cells and its mechanisms.Methods Cytotoxicity was analyzed by MTT.Early apoptotic cells stained with AnnexinV/PI were examined using flow cytometry.Protein expression was detected by Western blot analysis.Results Excisanin A inhibited proliferation of SW620 cells with IC_(50) value of 8.22 μmol·L~(-1) and induced cell apoptosis.Early apoptotic ratio increased rapidly within 12 h after 8.71 μmol·L~(-1) excisanin A treatment.Cleavage of the caspase-3,caspase-9 and caspase-3 substrate poly(ADP-ribose) polymerase(PARP) was observed in SW620 cells after treatment with different doses of excisanin A for 48 h.After SW620 cells were treated with 32.84 μmol·L~(-1) of excisanin A for different times,Western blot analysis showed stress-actived protein kinase(SAPK/JNK) and p38 mitogen-actived protein kinase were activated obviously after 3 h.Furthermore,c-Jun protein level was up-regulated after 6 h.Conclusions Excisanin A inhibited proliferation of SW620 cells and induced apoptosis of SW620 cells.Activation of SAPK/JNK and p38 pathways may be one of its mechanisms.
