Abstract
Objective: To study the effect of losartan on concentration of AngⅡand expression of c-jun mRNA in the left ventricle myocardium of rats with renovascular hypertension and to explore the possible mechanism of AngⅡreceptor1(AT 1) antagonists for reversion of left ventricle remoldeling. Method: Thirty ten-week-old male Wistar rats (weighting 190 to 220 g) were divided randomly into sham group (10) and 2KIC group(20). 2KIC group, left renal artery was partially obstructed by ligation with the residual diameter of 0.3 mm. sham group ,left and right renal artery were not touched, twenty renovascular hypertensive rats were divided randomly into losartan groupand control group. After eight weeks treatment , all animals were killed .Then the level of c-jun mRNA were measured by in situ hybridization, the AngⅡ in myocardium were measured by radioimmunoassay. Result: Afetr eight weeks teatment with losartan, the expression of c-jun mRNA decreased markedly (P 0.01), The AngⅡcontent in myocardium also decreased significantly by losartan (P 0.05). Conclusion: Losartan decrease content of AngⅡand expression of c-jun mRNA in the left ventricle myocardium of rats with renovascular hypertension, this may be one of mechanisms for AT 1 receptor antagonists to prevent and reverse left ventricle remodeling.
