Expression of TNF-α in the Ventricle Myocardium of Renovascular Hypertensive Rats

Abstract

Objective To observe expression of tumor necrosis factor-alpha(TNFα) in the ventricle myocardium of the renovascular hypertension rats.Methods A silver clip was placed on the left renal artery of Sprague-Dawley rats.After the operation the rats were divided into two groups: sham group and 2K1C(two-kidney one clip)group.Caudal vein blood pressure was measured every week.Animals were respectively killed after one,four,eight,twelve weeks.Then cardiac index(CI)was calculated,and TNF-α protein in ventricle myocardium was measured by ELISA and immunohistochemistry.Results The amount of myocardial TNF-α in 2K1C group was significantly higher than that in sham group.TNF-α increased significantly in the early development of renovascular hypertension,and it reached the peak when the renovascular hypertension stablized,and then declined.Conclusions TNF-α plays an important role during the formation and development of renovascular hypertensive.
