The influence of colchicine on glucose uptake by C6 glioma cells

Abstract

Objective To observe the influence of colchicine on glucose uptake by C6 glioma cells. Methods C6 glioma cells were incubated with colchicine for 3 h at 37℃, and then the cells were incubated in 1 ml PBS containing 125μmol/L unlabeled 2-deoxy-D-glucose and 37 × 103 Bq [3H]2-de-oxy-D-glucose for 5 min at 37℃. By using liquid scintillation analyzer, the glucose uptake by C6 glioma cells was determined. Results Colchicine could inhibit basal glucose uptake in a concentration-dependent manner by 7 % to 27 % . Glucose uptake was increased with carbachol by 11 % to 24 % , but this action was abolished by pretreatment with colchicines. Basal glucose uptake was increased by insulin in a concentration-dependent manner by 35 % to 47 %, but this action was not affected by pretreatment with colchicines. Conclusion Colchicine is involved in basal and carbachol-stimulated glucose uptake by C6 cells.
