Abstract

Objective To explore the relations between growth rate of glioma cell and glucose transport system.Methods C6 glioma cells were subcloned to produce C6 1,C6 2,C6 3 and C6 4.The level of GLUT1 mRNA and their protein of C6 1 cells and C6 4 cells were measured by Northern blot and Western blot respectively;at the same time,The rates of uptake of glucose by C6 4 cells and C6 1 cells were measured.Results The growth rates of the four cell clones differed from each other.The growth of C6 4 cells was about eight times faster than that of C6 1 cells.These clones predominantly expressed the GLUT1,the level of GLUT1 mRNA was markedly increased in C6 4 over C6 1 cells,densitometric analysis of the bands showed that the amount of GLUT1 protein in C6 4 cells was about three times higher than that in C6 1 cells [(310±21)%, P 0.001].The rates of uptake of 3 O methylglucose assayed by liquid scintillation count per minute:C64 cells was (7 680±180)/min·10 4cells,C6 1 cells was (3 000±125)/min·10 4cells;C6 4 cells was about 2.5 times higher than that by C6 1 cells.Conclusion These data suggest that the glucose transport system,coupled with glucose uptake,is closely related to the growth of glioma cells.
