Study on HPLC fingerprint of Radix Astragali from origination in Ningxia

Abstract

Objective To establish chromatographic fingerprint of Radix Astragali by HPLC which can be used to Radix Astragali's identity and standard planting.Methods A method of HPLC-UV was applied to establish chromatographic fingerprint of 16 Radix Astragali samples from different region and growth in Ningxia.The chromatographic condition:Thermo ODS-2HYPERSIL(250mm × 4.6mm,5μm) column,the mobile phase consisted of methanol,acetonitrile,and sodium acetate-acetic acid buffer(PH = 4.5) which was in the gradient elution at a flow rate of 1.0 mL.min-1,at 25℃.Results Established the chromatographic fingerprint of Radix Astragali in Ningxia,23 peaks were identified as the characteristic fingerprints of Radix Astragali.The similarity of Radix Astragali from different region and growth was calculated.Conclusion The similarity of Radix Astragali is related with its'region and growth.This fingerprint is useful for the identity and standard planting of Radix Astragali.
