Preparation of marine gastro-retentive sustained-release tablets and evaluation of its bioavailability in beagle dogs

Abstract

Objective:To prepare marine gastro-retentive sustained-release tablets and to evaluate its cumulative release ratio in vitro and pharmacokinetics in beagle dogs.Methods:Marine gastro-retentive sustained-release tablets were prepared with HPMC as matrix,cetyl alcohol as floating-assistant,magnesium carbonate as gas-producer.The dissolution of the tablets in 0.1 mol·L~(-1) HCl solution was measured.The plasma marine concentration in beagle dogs was measured by HPLC.The pharmacokinetic parameters of OMT in beagle dogs was evaluated and analyzed statistically by BAPP 2.0 program.The correlation of pharmacokinetics in vivo to in vitro was assessed using a Loo-Riegelman method.Results:The cumulative release ratio was eligible,and the tablets produced a relatively flat sustained plasma drug concentration in vivo compared with the marketing marine capsules.The relative bioavailability of the tablets was 110.5%.Conclusion:Marine gastro-retentive sustained-release tablets provided a sustained release of the drug.
