Abstract

Objective To study the alteration of TGF-β1 protein expression in astrocytes in vitro after mechanical injury,and get more messages of the repair mechanism of brain injury.Methods The astrocytes of cerebral cortex from SD rats which born in 24 hours were cultured in DMEM-F12 medium primarily.The cultured astrocytes randomly divided into control group and injury groups which were subjected to mechanical injury and then subdivided into 30min,1h,3h,6h,12h,24h,3d and 7d groups,the SABC immunohistochemistry method was used to examine the expression of TGF-β1 factor at different times after injury.Results The astrocytes cultured in vitro,more than 95% of the cells were astrocytes.There are no expression of TGF-β1 in the control groups.In the injury groups,the expression of TGF-β1 could be observed at 6h after injury,reached to the peak at 3d,then declined at 7d.Conclusion The expression of TGF-β1 after mechanical injury in vitro can be used as one of evidence for the timing of brain injury.
