The Study of S100β Expression in Rat Astrocytes in vitro after Mechanical Injury

Abstract

Objective:To study the alteration of S100β expression in astrocytes in vitro after mechanical injury,and get more messages of the repair mechanism of brain injury.Methods:The astrocytes of cerebral cortex from SD rats which was born in 24 hours were cultured in DMEM-F12 medium primarily.The cultured astrocytes were randomly divided into control group and injury groups which were subjected to mechanical injury and then subdivided into 30min,1h,3h,6h,12h,24h,3d and 7 d groups,the SABC immunohistochemistry method was used to examine the expression of S100β at different groups after injury.Results:More than 95% of the cells in the caltured medium were astrocytes.The low expression levels of S100β were observed in the control groups.In the injury groups,it could be detected increase in 30 min after injury,and increased significantly in 3~6h,and it reached apex 12h after injury,and decreased gradually to the level less than normal in 24h,and returned to normal in 7d.Conclusion:The expressions of S100β after mechanical injury in vitro is showed one-humped curve,but two-humped curve in vivo experiment.Though two kinds of curves are different,but both have time-dependent characteristic.It demonstrates that the expression of S100β after injury is one of evidence for inferring the timing of brain injury.The model of cell injury in vitro has superiority in studying the molecule mechanism of tissues or cell injury.
