Abstract

Objective To observe the scavenging effect on 3 kinds of different freedom radicals and the protective effect against the damage caused by hydrogen peroxide(H2O2) in endothelial cells of water extracts from honeysuckle flower and wild honeysuckle flower in different proportions(1∶ 0,2∶ 1,1∶ 1,1∶ 2,0∶ 1). Methods The scavenging ability of 5 different proportions of water extracts from honeysuckle flower and wild honeysuckle flower on 1,1-diphenyl-2-trinitro bitter hydrazine radical(DPPH·),hydroxyl radical(·OH),superoxide anion radical( O2-·) was determined to evaluate the antioxidant activity of the water extracts from honeysuckle flower and wild honeysuckle flower in different proportions. HUVECs were cultured in vitro to build the hypoxic-reoxygenation model,then the cell viability was determined by MTT,and the lactate dehydrogenase(LDH) and nitrogen monoxide(NO) level in cell culture medium of 5 kinds of different proportions of water extracts from honeysuckle flower and wild honeysuckle flower were detected. Results All of the different proportions had the antioxidant effect,and the honeysuckle flower and wild honeysuckle flower 2∶ 1 group had the best effect. All different proportions could effectively release the contention of NO and reduce LDH release from damaged cells,and the honeysuckle flower and wild honeysuckle flower 1∶ 0 group had the best effect,followed by honeysuckle flower and wild honeysuckle flower 2∶ 1 group. Conclusion All different proportions of water extracts from honeysuckle flower and wild honeysuckle flower have obviously anti free radical and antioxidant effects in vitro,and have protective effect on human umbilical vein endothelial cells injury,and the honeysuckle flower and wild honeysuckle flower 2∶ 1 group had the best effect.
