Abstract

16S rRNA gene clone library construction was used to analyze the microbial communities of soil sampled from above a known Ban876 gas reservoir.Two clone libraries DGS1 and DGS2 were constructed.There were 150 clones from the two libraries respectively for amplified ribosomal DNA restriction analysis(ARDRA).40 Taxanomic operational units(OTUs) were found in library DGS1,and 39 OTUs were found in library DGS2.Sequence analysis of the representative clone of each OTU showed that dominant bacteria of the DGS1 were affiliated with Gemmatimonadetes 28%,Chloroflexi 23%,Actinobacterium 21%;and Chloroflexi 19%,Sulfur-oxidizing 12%,Acidobacteria 10%,Gemmatimonadetes 10% for DGS2,which provides foundation for application of MPOG(Microbial prospecting of oil and gas).
