Abstract
Objecttive:to observe the acute and accumulate toxicity of nano-CuO to provide basis for clinic use.Methods:1.Acute toxicity test:activity condition and death condition were observed within fourteen days with different concentration of solutions of nano-CuO,and the method of krber was used to determine LD50 of nano-CuO and confidence level;2.Accumulate toxicity test:based on the accumulate toxicity of 20 days' design.50 of 20~22 g Kunming mice were divided into 5 groups randonmly.The first group was control group;28.90(1/20LD50)、57.81(1/10LD50)、115.62(1/5LD50)和298.05(1/2LD50)mg/(kg·bw) were added in the groups of 2,3,4and 5 through intragastric administration respectively in the form of nano-CuO,to test accumulate toxicity of nano-CuO.Results:1.Acute toxicity test:LD50 was 578.096 mg/(kg·bw) and confidence level was 343.92~971.63 mg/(kg·bw);2.Accumulate toxicity test:the results showed that the mice of the control and the first groups were all survived,the mice of the fifth group all dead;in the forth group there were half mice survived;in the third group there were 2 mice dead and 8 mice survived.Conclusion:high does of nano-CuO result in several mice kidney,intestina,liver and other organs damaged extensively,and it also caused organs serious congestion,hemorrhage and other pathological change.According to the standard,the test proved that nano-CuO had moderately accumulate toxicity to the mice.
