Abstract

【Objective】Caffeic acid-3-0-methyltransferase(COMT) is a key enzyme of lignin specific pathway.In this study,the role of GhCOMT1 gene in the cotton fiber development and thelignin specific biosynthesis pathway with transgenic technology was lucubrated.【Method】Vector of GhCOMT1 overexpress / anti-sense expression driven by RNAi specific promoter of cotton fiber E6 was constructed and transformaed into cotton.【Result】The GhCOMT1 gene has been cloned in every expression vector and h-arvesting transgenic offspring.【Conclusion】The function of GhCOMT1 gene was investigated,and the sense and anti-sense expression vectors of GhCOMT1 were constructed,in which the coding region of the gene was placed under the E6 prompter in either sense or anti-sense orientation.In addition,especial 450 bp fragments of GhCOMT1 NBD were also cloned,and this fragment was inserted into plant vector to construct RNAi expression vector of GhCOMT1.These constructed vectors were then transformed into Agrobacteri-um LBA4404 and successfully into cotton.
