Abstract
Objective To investigate the effect of melatonin on proliferation of cultured human retinal pigment epithelial cells (hRPECs), search for effective drug for the treatment of PVR. Methods Different concentration of melatonin (0,125 ng·L-1, 250 ng·L-1, 500 ng· L-1) were added to hRPECs respectively , after 72 hours using MTT method at automatic enzyme labeling machine value of A were calculated at 540 nm and inhibition rate of retinal pigment epithelial(RPE) cells were measured by MTT. Results Melatonin at different concentration could inhibit RPE cells in dose dependent manner, in comparision of different experimental group 125 ng·L-1 and 500 ng·L-1 with control group difference was highly significant(P 0.001) ,but comparision of experimental group 250 ng· L-1 with control difference was not significant. Conclusion Melatonin can effectively inhibits hRPECs growth in vitro, and it might be a new candidiate drug of preventing for patients with proliferative vitreoretinopathy.
