Effects of Atorvastatin on matrix metalloproteinase-1 secreting by rat vascular smooth muscle cells

Abstract

Objective To investigate the effects of atorvastatin on matrix metalloproteinase-1(MMP-1) secreting by rat vascular smooth muscle cells.Methods Rat aortal smooth muscle cells were first incubated in serum-free medium with 10 ng/ml IL-1β;24 hours later,atorvastatins with different concentration(1×10-7mmol/L,1×10-6mmol/L,1×10-5mmol/L) were added to the medium.48 hours later,supernatants of different cultures were collected and MMP-1 levels were determined by ELISA kits according to the manufacturer's instructions.In the group with atorvastatin concentration of 1×10-5mmol/L,MMP-1 levels were respectively determined after 6,24 and 48 hours.Results Atorvastatin inhibited the secretion of MMP-1 in a time-dose-dependent manner.Conclusions Atorvastatin can inhibit MMP-1 secreting by IL-1β stimulated rat vascular smooth muscle cells.
