Separation and Purification Process of Total Flavones from Radix tetrastigmae with Macroporous Resins

Abstract

OBJECTIVE To screen the suitable resin for separation and purification of total flavones from Radix tetrastigmae,and to establish the optimum parameters of separation and purification process.METHODS The adsorption capacities and desorption ratios of ten selected macroporous resins were evaluated with UV method.Suitable resins were chosen to study the performance of static-dynamic adsorption and desorption,and to optimize the process parameters.RESULTS HPD826 resin showed advantageous adsorption and desorption properties with thermodynamic parameter according with the Freundlich equation.The optimal conditions were determined as follows: for adsorption,the flow speed was 2 BV·h-1;for desorption,wash the adsorbate-laden column first with 2 BV water,then with 3 BV 70% ethanol-water.Under this condition,the content of total flavones increased by 2.44-fold.CONCLUSION It is feasible to separate and purify Radix tetrastigmae total flavones with HPD826 resin.
