Abstract
Objective To explore the expression and significance of Nanog in gliomas of different pathological grades.Methods The expression of Nanog mRNA was examined by reverse transcription polymerase chain reaction (RT-PCR) and Nanog protein was detected by immunohistochemistry in the specimens of 50 cases of glioma tissues and 7 cases of normal brain tissues.Results The immunopositive ratio was 18.3%±9.6% in WHO Grade Ⅱ,37.9%±19.0% in WHO Grade Ⅲ,53.4%±19.8% in WHO Grade Ⅳ,respectively (F=14.918,P=0.000).The relative mRNA level of Nanog was 0.1824±0.0310 in WHO Grade Ⅱ,0.3730±0.0961 in WHO Grade Ⅲ,0.4594±0.0453 in WHO Grade Ⅳ,respectively (F=65.901,P=0.000).The relative expression level of Nanog obviously correlated with the pathological grades.The Nanog protein and mRNA were not detected in normal brain tissues.Conclusion Nanog is overexpressed in gliomas.The relative expression level of Nanog is related to the pathological grades,which lays a foundation to the further research on its relation with brain tumor stem cells and its role in biological behaviors of gliomas.
