Effect of spironolactone on aldosterone-induced plasminogen activator inhibitor-1 expression of rat glomerular mesangial cells

Abstract

Objective To investigate the role of spironolactone on aldosterone-induced plasminogen activator inhibitor-1(PAI-1)expression of rat glomerular mesangial cells.Methods The rat mesangial cells were cultured in the medium with normal glucose,10-7 mol·L-1 aldosterone and different concentrations of spironolactone(10-7,10-8,10-9 mol·L-1).RT-PCR was used to detect the expression of PAI-1,mineralocorticoid receptor(MR) and 11β-hydroxysteroid dehydrogenase(11β-HSD2) mRNAs.The levels of PAI-1 in the supernatants were determined by ELISA.Results MR and 11β-HSD2 mRNA expressions were detectable in mesangial cells.Compared with normal glucose group,PAI-1 mRNA expression of mesangial cells and PAI-1 level in the supernatants increased significantly in aldosterone group,while spironolactone inhibited those changes in a dose-dependent manner.Conclusion Spironolactone,which compete to bind with MR against aldosterone,can ameliorate aldosterone-induced PAI-1 expression in mesangial cells.
