Abstract
Objective To investigate the effects of resveratrol on the expression of PDCD5 of 786-0 human kidney cancer cells.Methods Appling the resveratrol of different density(12.5 μmol/L,25 μmol/L,50 μmol/L,100 μmol/L) affects on 786-0 human renal cell 24h.The effect of Resveratrol on the expression of apoptosis-related protein PDCD5 was analyzed by using Immunocyto chemical detection of SP method;the cell cycle phase distribution and the cell apoptosis of human kidney cancer 786-0 cells were analyzed by using flow cytometry.Results The proliferation of RCC 786-0 cell was inhibited in a dose-dependent by resveratrol.The level of expression of human renal cancer 786-0 cells PDCD5 was low,but the level was significantly higher after dealling with RES;Compared with control group,P0.05.The resveratrol treatment groups and the control group compares,the difference has statistics significance(P0.05).The experiment groups compareing with each other,the 12.5 μmol /L group with 25 μmol /L group、50 μmol/L group、100 μmol/L group have a significant difference(P0.05);but the 25 μmol/ L group and 50 μmol /L group,100 μmol/L group are not statistically significant(P0.05).Flow cytometric analyses showed that 786-0 cells were obviously inhibited,the ratio of 786-0 cells in the S phase were apparently increased at 24h resveratrol treatment of different concentrations.Conclusion The expression of PDCD5 of 786-0 human renal cell carcinoma cells can be increased by RES.786-0 human renal cancer cells has been arrested in S phase and induce apoptosis by resveratrol,and this effect has a dose-dependent.
