Abstract
Objective To investigate the effects of bufalin on proliferation and apoptosis of cisplatin-resistant gastric cancer cell(SGC7901/CDDP) in vitro and the underlying mechanisms.Methods Cell proliferation was measured using MTT assay and cell apoptosis was determined by flow cytometry after propidium iodide staining.The expressions of Bcl-2 and Bax proteins were detected by Western blot.Results Bufalin could inhibit the proliferation of SGC7901/CDDP in the time-and dose-dependent manners.The concentration of inhibited cell viability(IC50) for 24,48 and 72 h was 106.3,53.8 and 10.9 nmol/L,respectively.After treatment with bufalin of 10,50 and 100 nmol/L for 24 h,the rate of SGC7901/CDDP apoptosis was 7.3%,12.6% and 26.4%,respectively,which was concentration-dependent.When the cell was treated with bufalin of different concentrations,the expression of Bax was up-regulated but the expression of Bcl-2 was down-regulated.Conclusion Bufalin can inhibit the proliferation of cisplatin-resistant SGC7901/CDDP.The cell apoptosis is related to the up-regulated expression of Bax and the down-regulated expression of Bcl-2.
