Abstract
OBJECTIVE To establish a way for extracting and separating solanesol from tobacco leaves. METHODS The extraction rate and the content of solanesol was determined by HPLC and the extraction rate and the content of solanesol were used as quality control criteria. The effect of different parameters such as extraction time,temperature and pressure on the supercritical carbon dioxide extraction of solanesol was investigated. RESULTS The optimal extraction conditions were determined according to orthogonal experiments as entrainer with 95% ethanol (volume fraction),extraction pressure at 20 MPa,extraction temperature at 45 ℃ and extraction time for 3 h. The extraction rate of solanesol was 94.5% and the content of solanesol was 33.8%.The content of purified sample was 93.4%. CONCLUSION This technology was feasible .The purity coefficient and yield of solanesol extracted by SFE and separated by column chromatograph were high.
