Inhibitor LY294002 of PI3K/AKT signal pathway mediated by ICBP90 inhibits proliferation of Jurkat T cell

Abstract
Aim:To investigate the role of LY294002 which is the inhibitor of PI3K/Akt signal pathway in proliferation of Jurkat T and expression level of ICBP90. Methods: Jurkat T cells were used as a suitable model for this study.The positive control was Adriamycin and PD98059.Inverted microscope was used to observe the colony formation of Jurkat T cells.The proliferation of cells treated by LY294002,PD98059,Adriamycin,LY294002 combining with Adriamycin and PD98059 combining with Adriamycin,respectively,was checked by MTT method.The effect of these drugs on the cell cycle distribution of Jurkat T cells was analyzed by flow cytometry.Western blotting was used to determine the level of ICBP90 expression in Jurkat T cells treated by the drugs.Results: The proliferation of Jurkat T cells was inhibited by LY294002.LY294002 can make Jurkat T cell arrest at G2/M phase.The expression of ICBP90 declined in the group added LY294002.LY294002 combining with Adriamycin exerted the synergetic role to some extent.Conclusion:LY294002 can down-regulate expression of ICBP90 and inhibit Jurkat T cell proliferation,indicating that ICBP90 might mediate LY294002 to inhibit Jurkat T cell proliferation.
