γ-secretase inhibitor DAPT suppresses the proliferation of Jurkat T cells

Abstract

Aim:To explore effects of γ-secretase inhibitor DAPT on the proliferation of Jurkat T cells and its mechanism in vitro. Methods: The colony formation of Jurkat T cells was observed under an inverted microscope and the proliferation of them was detected by MTT assay.Then,the cell cycle distribution of Jurkat T cells induced by DAPT was analysed by flow cytometry combining with propidium iodide staining.The expression of ICBP90 in Jurkat T cells was detected by Western blotting.Results: DAPT significantly inhibited the colony formation and the proliferation of Jurkat T cells in a dose-dependent manner,and caused S phase arrest.DAPT treatment induced a decrease in the expression level of ICBP90 protein.Conclusion:the inhibitor of γ-secretase,DAPT,may inhibit the proliferation of Jurkat T cells and their cell cycle,which is related to the decrease of ICBP90.
