Simultaneous determination of paeoniflorin and albiflorin in Radix Paeoniae Alba by UPLC

Abstract

Objective To establish a UPLC method for the simultaneous determination of paeoniflorin and albiflorin in Radix Paeoniae Alba.Methods Agilent Extend-C18(4.6 mm×50 mm,1.8 μm) was used with acetonitrile(A)-0.2% acetic acid solution(B)as the mobile phase in gradient elution[0-2.5 min,8%-12%A;2.5-10 min,12%-20%A].The flow rate was 0.5 mL·min-1,the column temperature was 30℃,and the detection wavelength was 230 nm.Results The linear ranges of paeoniflorin and albiflorin were 0.011 6-1.45(r=0.999 9) and 0.008-1 mg·mL-1(r=0.999 9);the average recoveries were 98.6% and 97.5%;and the RSDs were 1.4% and 2.2%.Conclusion The method is rapid,sensitive,and reliable,which can be used for the quality control of Radix Paeoniae Alba.
