Abstract

Objective:To establish a high performance liquid chromatography method for the determination of paeoniflorin and albiflorin in Radix paeoniae alba.Methods:The HPLC method for determination of paeoniflorin and albiflorin was:Kromasil ODS2 column:(4.6mm×250mm,5μm),mobile phase:methanol-0.1% phosphate solution(adjusted to pH 4.2 with 1% sodium hydroxide)(34:66),flow rate:1.0 mL?min-1,detector wavelength:230 nm.Results:The linear range of paeoniflorin and albiflorin was 0.028~0.896μg(r =0.9999) and 0.0359~0.574μg(r = 0.9999),The average recovery was 98.18 % and 98.26% respectively,and both the RSD(n = 9) were less than 2%,The content determination of radix paeoniae alba of different producing areas and batches indicated that the content of paeoniflorin no less than 2.71%,albiflorin no less than 0.75%.Respectively,the average values was 2.93% and 0.95%.Conclusion:The method is sensitive,accurate,simple,and it can determine the paeoniflorin and albiflorin in radix paeoniae alba simultaneously.
