HPLC quantitative analysis of four flavonoids in crude baicalin

Abstract

OBJECTIVE To establish an HPLC method for determination of baicalin,baicalein,wogonoside and wogonin,in crude baicalin.METHODS Using Agilent ZORBAX SB-C18 column(5 μm,4.6 mm×250 mm).The mobile phases for glycosides consisted of methanol-water-acetic acid(49∶51∶0.2).The mobile phases for aglycones consisted of methanol-water-acetic acid(61∶39∶0.2).The UV detector at 275 nm.RESULTS The linear ranges of baicalin,baicalein,wogonoside and wogonin were 0.102-1.020 μg(r=0.999 7),0.051-0.510 μg(r=0.999 4),0.01-0.51 μg(r=0.999 5),0.02-0.20 μg(r=0.999 9),respectively.The average recoveries(n=5) of baicalin,baicalein,wogonoside and wogonin were 99.12%(RSD =1.55%),101.60%(RSD =2.67%),102.27%(RSD =1.92%)and 98.51%(RSD=3.13%),respectively.CONCLUSION The method is simple,accurate and can be used to determine the contents of crude baicalin.
