Effect of Bushen Huoxue Recipe on the Expression of p16 Gene of Vascular Endothelial Cells in Rats

Objective To investigate the effect of Bushen Huoxue Recipe on the expression of p16 gene of vascular endothelial cells in rats. Methods A total of 24 healthy male SD rats were randomly divided into groups A,B,C,8 rats in each group. A group given Bushen Huoxue Recipe,B group given Astragalus Membranaceus,C group given 0. 9% sodium chloride solution,the dosage of each group was equal,and the drug- delivery way was gavage. Human umbilical vein endothelial cells were cultivated by medicated serum,β- galactosidase staining was used to authenticate cell senescence,flow cytometry was used to analyze the changes of cell cycle,and RT- PCR method was used for the detection of expression of p16 gene. Results G0/ G1 cell proportion of groups A and B was lower than that of C group,respectively,and that of A group was lower than that of B group( P 0. 05). Expression of p16 gene of groups A and B was lower than that of C group,respectively,and that of A group was lower than that of B group( P 0. 05). Conclusion Bushen Huoxue Recipe can delay the senescence of vascular endothelial cells in rats,its molecular mechanism may be regulate expression of p16 gene to make cell cycle stagnated at G1 phase.
