Determination of organic acids in succinic acid fernmentation broth by reversed phase high performance liquid chromatography

Abstract

For the detectation of succinic acid fermentation, a reversed phase high performance liquid chromatography method on a column of synergi Hydro-RP C18(4.6 mm×250 mm, 4 μm) with a eluent containing methanol and 0.01 mol/L potassium dihydrogen phosphate buffer(pH2.0)(5 ∶95, v/v) was used, the flow rate of mobile phase was 1.0 mL/min, the detection wavelength was 215 nm and the column temperature was room temperature. Before sampling, the fermentation broth was centrifuged and filtered. Succinic acid, formic acid, acetic acid and fumaric acid were compeletly separated and determined in 10 minutes. The recovery and relative standard deviation(RSD, n=5) of organic acid were from 98.18% to 101.28%, in the range of 1.24%~4.87%, respectively. The mothod was proved to be a rapid, effective way to determine the organic acid in succinic acid fermentation broth.
