Abstract
Objective:To study the expression of GLUT1 and GLUT4in placenta tissue of gestational diabetes mellitus patients and its relationship with related miRNAs.Methods:Placental tissues were collected from 30 cases of gestational diabetes mellitus puerperas and 30 healthy puerperas and then fixed by formalin and frozen in liquid nitrogen respectively;then translation levels of GLUT1 and GLUT4were detected by immunohistochemistry method,and transcription levels of GLUT1 and GLUT4as well as expression of miRNA-518 dwere detected by fluorescent quantitative PCR.Results:(1)mRNA contents and the mean optical density of GLUT1 and GLUT4in placental tissue of GDM group were lower than those of control group;(2)expression of miRNA-518 din placental tissue of GDM group was higher than that of control group;(3)target scan software forecast showed that there were binding sites of miRNA-518 din 3'UTR of mRNA of GLUT1 and GLUT4;(4)miR-518 content was negatively correlated with mRNA contents and protein contents of GLUT1 and GLUT4.Conclusion:Expression of GLUT1 and GLUT4decreases but expression of miRNA-518 dincreases in placental tissue of gestational diabetes mellitus;miRNA-518 dmay affect glucose transport and insulin sensitivity by inhibiting the expression of GLUT1 and GLUT4.
