Abstract
Objective To investigate the inhibitory effects of rapamycin on proliferation and migration of human umbilical vein endothelial cells(HUVECs) in vitro.Methods Using MTT assay,flow cytometry and migration assay,the effects of rapamycin with different concentration on the proliferation of HUVECs stimulated by VEGF were evaluated.Results Rapamycin inhibited proliferation of HUVECs stimulated by VEGF at the concentration of 10 ng/ml after 48 h.Flow cytometry analyses revealed that rapamycin treatment arrested HUVECs at the G0/G1 phase of the cell cycle and induced apoptosis of HUVECs;The inhibition on migration of HUVECs in response to VEGF was observed in the presence of 10 ng/ml rapamycin.Conclusions Rapamycin inhibited the proliferation and migration of vascular endothelial cells may contribute to anti-angiogenesis.
