Influence of Xiaoyao powder on expression of NR2A and NR2B in brain NMDA receptor by chronic restraint stress

Abstract

Objective To observe the change of protein expressions of NR2A and NR2B in N-methyl-D-aspartic(NMDA) receptor in hippocampus and basolateral lateral amygdaloid(BLA) on restraint stress as well as the regulation action of Xiaoyao powder.Methods With tie-up method to establish animal model of chronic restraint stress,Xiaoyao powder was applied to intervene.The expression status of proteins in CA1,CA3,dentate gyrus(DG) of hippocampus,and NR2A,NR2B in NMDA receptor of BLA were tested after 7 and 21 days respectively.Results There was expression of NR2A and NR2B to varying degrees in CA1,CA3,DG of hippocampus and BLA.The expression of NR2A and NR2B in the Xiaoyao powder group on Day 21 was all increased in CA1,CA3,DG and BLA(P 0.05,P 0.01),which was especially obvidus in CA3 and DG of hippocampus(P 0.01).Conclusion The Xiaoyao powder has obvious regulation action on chronic restraint stress-induced neurotransmission change in hippocampus and BLA,which could make an influence on plasticity of synapsis to satisfy body function,and the main target is CA3 and DG in hippocampus.
