Abstract
Objective To study the effect of high glucose on cultured vascular endothelial cells.Methods Cultured vascular endothelial cells(ECV304) were treated with various concentrations of glucose for 24-72 hours.The inhibition of proliferation was measured by using MTT assay.Phase contrast microscope and transmission electron microscope were employed to observe cell apoptosis.The apoptosis rate in ECV304 was measured by using flow cytometer.Results High glucose significantly inhibited ECV304 proliferation(P0.01)and induce apoptosis in a dose and time dependent manner than the low-glucose group(P0.01).Conclusion High glucose could inhibit the proliferation and accelerate cell apoptosis of ECV304.
