Abstract
Objective To study the effects of different cell preparation methods on proportion of cytokine-induced killer(CIK) cell subsets.Methods Forty-two CIK cell samples were equally randomized into two groups.Group A was prepared according to the conventional method and group B was treated with recombinant human granulocyte colony-stimulating factor(rhG-CSF) 10 ng/ml on the 2nd day on the basis of conventional method.Flow cytometry was used to detect the proportion of CIK cell subsets after cell maturation in two groups.Results Compared with group A,the proportion of CD3+CD4+ T cell was decreased(P0.01),which of CD3+CD56+ T cell was increased(P0.05) in group B.There was no significant difference in the proportion of CD3+CD8+ T cell between two groups(P0.05).Conclusion rhG-CSF may inhibit the induction of CD3+CD4+ T cells and improve the induction of CD3+CD56+ T cells in the preparation of CIK cells.
