Abstract
Objective To observe the effect of Allicin on TRAIL-induced apoptosis in ovarian cancer SKOV-3 cells in vitro,and to explore its action mechanism.Methods Human ovarian cancer cell lines SKOV-3 in vitro were treated with Allicin and TRAIL respectively or collectively.The cell growth inhibition rates were detected by MTT assay.The expressions of DR4 and DR5 were detected by FCM.The activities of Caspase 3,8 were detected by spectrophotometer.Results(1)The inhibitory rates for SKOV-3 cells at 24,48,72 h after treated by Allicin and TRAIL respectively or collectively showed a time-and-dose-dependent manner(P0.05).(2)The expressions of DR4 and DR5 were upregulated significantly 24h after treated with Allicin.FI was increased from 1.78 to 2.27 for DR4 and from 1.94 to 2.58 for DR5(P0.05).(3)The activities of Caspase-3 and Caspase 8 were upregulated obviously 48h after treated with Allicin.The ratio of OD of Allicin group vs blank group of Caspase-3,8 was 2.49,2.08 times of that in control group respectively(P0.05).Conclusion(1)The ovarian cancer cell line SKOV-3 is sensitive to Allicin and TRAIL.(2)Allicin can increase TRAIL-induced apoptosis in ovarian cancer SKOV-3 cells.(3)Allicin can upregulate TRAIL death receptor 4,5 and increase Caspase-3,8 activity,which may be its action mechanism of enhancing TRAIL-induced apoptosis.
