Abstract

PurposeTo study influential factors of the high density fermentation of recombinant human TNF-related apoptosis-inducing ligand(TRAIL) Pichia pastoris.MethodsThe effects of different methanol inducement period, methanol concentration,pH were observed using shaking flask, and fed-batch fermentor was also studied.Results72.8 mg/L rhsTRAIL was obtained at pH 6.0,1.0% methanol on the fourth day and 184 mg/L rhsTRAIL were obtained in fed-batch fermentor.ConclusionThe developed fermentative technology might incease the expression of sTRAIL in recombinant Pichia pastoris strain.
