Effect of PAMD on Bcl-2 mRNA and Bax mRNA expression following cerebral ischemia

Abstract

To explore the expressions of Bcl-2 mRNA and Bax mRNA in the cortical neuron induced after ischemia modulated by PAMD.The rat cerebral ischemia models are made by middle cerebral artery occlusion(MCAO) using improvement methods introduced by Zea Longa.The rats are pretreated with PAMD for 7 days before ischemian in experimental drug pretreatment group.The changes of Bcl-2 mRNA and Bax mRNA expressions in the cerebral cortex of rats are measured by reverse transcription-polymerase chain reaction(RT-PCR) method in shame operation group,model group and PAMD high dose,middle dose,low dose,Nimodipine and ginkgo leaf tablet pretreatment group at 24 hours after ischemia. After MCAO,Bcl-2 mRNA and Bax mRNA are both induced to express.Level of Bcl-2 mRNA decreased,while the level of Bax mRNA increased.In comparison with the model group,after PAMD pretreatment for 7 days,Bcl-2 mRNA in the cerebral cortex is up-regulated at 24 hours after ischemia while Bax mRNA is down-regulated at 24 hours after ischemia.PAMD can efficiently regulate Bcl-2 mRNA and Bax mRNA expressions in cortical neuron to protect the nerves after cerebral ischemic injury of rats.
