Effect of cyclosporin A on activation of immune cells in brain after chronic cerebral hypoperfusion in aged rats

Abstract

Objective To explore effect of immunosuppressant cyclosproin A(CsA) on the activation of immune cells in brain after chronic cerebral hypoperfusion(CCH) in rats. Method Both common carotid arteries were ligated bilaterally in 50 Wistar rats. Twelve of them were filled CsA. The activation of immune cells was monitored by immunohistochemical methods. The histopathologic damages of brain were observed with optical microscope. The study was then investigated for 2 months after the CCH. Result There were extensive activtion of microglia and blood- derived T- cells and leukocytes , and histopathologic damages of brain after CCH. In CsA treated rats, the activation of immune cells in brain was significantly suppressed, histopathologic damages of brain were much less intense also.Conclusion There was an extensive activtion of immune cells in brain after CCH. CsA can suppress evidently acivation of them and reduce histopathologic damages of brain.
