Abstract
Objective To inquire into the cerebral gene changes in mice with kidney-yang deficiency caused by overwork and intemperance in sexual activity following the treatment of Jinguishenqi Pills(JP) or Yougui Pills(YP).Methods Forty-five male mice were divided into 4 groups,with 10 in the control group,15 in the model group,10 in the JP group and 10 in the YP group.Every mouse in the control and model group was given distill water 0.5 mL per day,and every mouse in the JP and YP group were given 0.5 mL suspension of JP or YP per day.The male mice in the model,JP and YP group lived in the same cage with female mice at the ratio of 1∶ 6(male:female),and swam 30~40 min everyday for 4 weeks to result in kidney-yang deficiency caused by overwork and intemperance in sexual activity.The brain gene expression of the mice were detected with mouse brain gene chip of 36K mouse genome array made by CapitalBio Corp.Beijing,China.Results Compared with the control group,186 differential expression genes were screened in the model group,including 150 up-regulated genes and 36 down-genes;299 differential expression genes were screened in the JP group,including 114 up-regulated genes and 185 down-regulated genes;155 differential expression genes were screened in the JP group,including 64 up-regulated genes and 91 down-regulated genes.There were 24 up-regulated genes both in the JP and YP group,accounting for 21.05 percent and 37.5 percent in each group respectively,which were mainly the genes related to neurotransmissions/signal transduction,transcription/translation,cellular cycle,hormone and metabolism.The down-regulated genes both in the GP and YP group were 35,accounting for 18.92 percent and 38.46 percent in each group respectively,chiefly including the genes associated with inflammation/immunization,neurotransmissions/signal transduction,transcription/translation,metabolism and limiting velocity enzyme of dopamine.Compared with the control group and model group respectively,there were 23 genes up-regulating in the model group while down-regulating in the treatment group,mainly involving the genes related to inflammation/immunization and neurotransmissions/signal transduction.There were six genes down-regulating in the model group,while up-regulating in the treatment group,mainly including the related genes of cellular cycle and structure,neurotransmissions/signal transduction and transcription.Conclusion JP and YP may down-regulate the expressions of the genes related to hormone and melanin-concentrating hormone(MCH) which are up-regulated in mice with kidney-yang deficiency.This study can inquire into the effect mechanism of the two drugs and their differences in genetic level.
