Extraction of Salvianolic Acid B from Danshen (Salvia Miltiorrhiza Bge. ) and Determination by HPLC Method

Abstract

Objective To optimize the conditions for extracting Salvianolic acid B from Danshen (Salvia miltiorrhiza Bge. ). Methods Danshen crude drugs were dried, ground and sieved through 60-mesh screen sieve, followed by 20min ultrasonication in ice bath with different solvents. The Danshen suspensions were filtered and Salvianolic acid B in the filtrate was determined by HPLC. The column used was Alltima C18 (5μm, 250 mm×4. 6 mm); The mobile phase was with acetonitrile-0. 5% acetic acid solution in program of elution; the flow rate was 1. 0 mL/min and the detect wavelength was 290 nm. Results With ice bath, 70% methol achieved the highest rate of extraction, and with-water extraction was 95. 06% of that with 70% methol. Repeatability and precision were good. Salvianolic acid B had a good linearity within the range of 0. 07-0. 70μg (r=0. 9997),and the RSD was less than 3%. Conclusion The treatment of samples was simple and the results were accurate. This method may be used for extraction and determination of Salvianolic acid B.
